Description

Description: This assay employs a two-site sandwich ELISA to quantitate ZGPAT in samples. An antibody specific for ZGPAT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ZGPAT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZGPAT is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZGPAT bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: ZGPAT, Contains 1 C3H1-type zinc finger. Contains 1 G-patch domain. Transcription repressor that specifically binds the 5'-GGAG[GA]A[GA]A-3' consensus sequence. Represses transcription by recruiting the chromatin multiprotein complex NuRD to target promoters. Negatively regulates expression of EGFR, a gene involved in cell proliferation, survival and migration. Its ability to repress genes of the EGFR pathway suggest it may act as a tumor suppressor. Able to suppress breast carcinogenesis. Six new targets of epigenetic inactivation (IRX3, WNT10A, WNT6, RARalpha, BMP7 and ZGPAT). These targets displayed cell line and tumor specific differential methylation when compared with normal brain samples, suggesting they may have utility as biomarkers
